چکیده

We recently reported (Clewell et al., 1972) on an inhibitory effect of rifampicin on Co1 E 1 plasmid replication. The present study represents a further characterization of this phenomenon as well as a study of the effects of two other known inhibitors of RNA synthesis, streptolydigin and actinomycin D. During treatment of cells with chloramphenicol the colicinogenic factor El (Co1 El) continues to replicate for more than ten hours. During this time 4 to 5 S RNA is also synthesized. When varying concentrations of rifampicin were inchIded during chloramphenicol treatment, inhibition of plasmid DNA synthesis correlated very closely with inhibition of cellular RNA synthesis. Similar experiments testing the effects of streptolydigin and actinomycin D (during chloramphenicol treatment) showed that cellular RNA synthesis was at least 100 times more sensitive to these drugs than was plasmid DNA synthesis. When actively growing cells (i.e. cells not treated with chloramphenicol) were treated with a high concentration of rifampicin (250 pg/ml), chromosomal DNA synthesis continued to an extent representing about a 50% increase in DNA, while plasmid DNA synthesis appeared to stop abruptly.

Segregation into and replication of plasmid deoxyribonucleic acid in chromosomeless segregants of Escherichia coli.

The progeny cells of Escherichia coli strain P678-54, which normally do not contain deoxyribonucleic acid (DNA), were found to carry DNA when the parental cell carried colicin factor E1 (Col E1). The DNA found in the normally DNA-less segregants was shown to be Col E1 DNA, which is present primarily as a covalently closed circular molecule that can undergo more than one complete cycle of replic...

Discontinuous replication of colicin E1 plasmid deoxyribonucleic acid.

The plasmid pML 21, which was found to contain approximately 49% of the Col E1 genome was used to determine the template origin of single-stranded deoxyribonucleic acid (DNA) fragments (4 to 32% of the Col E1 units length) associated with Col E1 dna replicative intermediates. The results of DNA hybridization competition experiments indicate that the single-stranded fragments derive from the ful...

Utilization of two distinct modes of replication by a hybrid plasmid constructed in vitro from separate replicons.

A hybrid plasmid, pSC134, that codes for two distinct sets of replication functions has been constructed in vitro by ligation of EcoRI endonuclease-cleaved pSC101 and Col E1 plasmid replicons, and has been introduced into Escherichia coli by transformation. The replication properties of the pSC134 plasmid in DNA polymerase I-defective mutants or in the presence of chloramphenicol indicate that ...

On the mechanism of streptolydigin inhibition of Escherichia coli RNA polymerase.

The mechanism of streptolydigin inhibition of RNA synthesis has been investigated with a combination of steady state kinetics and product analysis by employing the abortive initiation reaction of Escherichia coli RNA polymerase. The pattern of inhibition by streptolydigin on the poly[d(A-T)] . poly[d(A-T)]template (non-competitive versus AMP; competitive versus UTP) was consistent with one inhi...

Replication of mini-P1 plasmid DNA in vitro requires two initiation proteins, encoded by the repA gene of phage P1 and the dnaA gene of Escherichia coli.

We have developed an in vitro DNA-replication system that replicates exogenously added mini-P1 plasmid DNA. The system consists of purified P1 RepA protein and a partially purified mixture of Escherichia coli replication proteins. It is essentially the same as that described for the replication of oriC plasmid DNA [Fuller, R.S., Kaguni, J.M. & Kornberg, A. (1981) Proc. Natl. Acad. Sci. USA 78, ...
